Pollen analysis explained
Melissopalynology — counting and identifying the pollen grains in a honey under a microscope. It is how monofloral claims are substantiated, how geographic origin is challenged, and it is far less straightforward than it sounds.
How it is done
A weighed sample of honey is dissolved in warm water and centrifuged. The sediment — pollen grains, plus yeast, algae, honeydew elements and whatever else was suspended in the honey — is mounted on a slide and examined at high magnification. An analyst identifies grains by their shape, surface sculpture and aperture pattern, and counts several hundred to establish proportions.
It is slow, skilled work. Identification depends on a reference collection and on an analyst who knows the regional flora, and two competent laboratories can return meaningfully different results on the same jar. That is not a scandal; it is the nature of a method that rests on visual identification of objects a few tens of micrometres across.
Why the raw percentage misleads
Pollen does not enter honey in proportion to nectar. It is carried in on the bee's body and shaken loose, so a plant that produces abundant pollen is over-represented and a plant that produces little is under-represented — sometimes by an order of magnitude. Correction factors exist for the well-studied sources, and applying them is what separates analysis from grain-counting.

Pollen as a geographic witness
The secondary and minor pollens are often more informative than the dominant one. A honey sold as European whose slide carries pollen from plants that do not grow in Europe has answered a question about origin that no sugar test could. This is one of the few practical checks on origin fraud, and it is used in enforcement.
